NM_001103.4(ACTN2):c.58A>G (p.Met20Val) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.M20V variant (also known as c.58A>G), located in coding exon 1 of the ACTN2 gene, results from an A to G substitution at nucleotide position 58. The methionine at codon 20 is replaced by valine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
