Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000051.4(ATM):c.5875G>A (p.Glu1959Lys), citing Ambry Variant Classification Scheme 2023: The p.E1959K variant (also known as c.5875G>A), located in coding exon 38 of the ATM gene, results from a G to A substitution at nucleotide position 5875. The glutamic acid at codon 1959 is replaced by lysine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
