Uncertain significance — the classification assigned by Ambry Genetics to NM_021930.6(RINT1):c.580A>G (p.Met194Val), citing Ambry Variant Classification Scheme 2023: The p.M194V variant (also known as c.580A>G), located in coding exon 5 of the RINT1 gene, results from an A to G substitution at nucleotide position 580. The methionine at codon 194 is replaced by valine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
