Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007294.4(BRCA1):c.572T>C (p.Val191Ala), citing Ambry Variant Classification Scheme 2023: The p.V191A variant (also known as c.572T>C), located in coding exon 7 of the BRCA1 gene, results from a T to C substitution at nucleotide position 572. The valine at codon 191 is replaced by alanine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
