Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000138.5(FBN1):c.572G>T (p.Ser191Ile), citing Ambry Variant Classification Scheme 2023: The p.S191I variant (also known as c.572G>T), located in coding exon 6 of the FBN1 gene, results from a G to T substitution at nucleotide position 572. The serine at codon 191 is replaced by isoleucine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
