Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001130438.3(SPTAN1):c.5701G>A (p.Glu1901Lys), citing Ambry Variant Classification Scheme 2023: The p.E1901K variant (also known as c.5701G>A), located in coding exon 43 of the SPTAN1 gene, results from a G to A substitution at nucleotide position 5701. The glutamic acid at codon 1901 is replaced by lysine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
