Uncertain significance — the classification assigned by Ambry Genetics to NM_003579.4(RAD54L):c.56C>G (p.Ser19Cys), citing Ambry Variant Classification Scheme 2023: The p.S19C variant (also known as c.56C>G), located in coding exon 2 of the RAD54L gene, results from a C to G substitution at nucleotide position 56. The serine at codon 19 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
