Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_005585.5(SMAD6):c.565G>C (p.Asp189His), citing Ambry Variant Classification Scheme 2023: The p.D189H variant (also known as c.565G>C), located in coding exon 1 of the SMAD6 gene, results from a G to C substitution at nucleotide position 565. The aspartic acid at codon 189 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
