NM_001277115.2(DNAH11):c.55C>T (p.Leu19Phe) was classified as Uncertain significance for Primary ciliary dyskinesia by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L19F variant (also known as c.55C>T), located in coding exon 1 of the DNAH11 gene, results from a C to T substitution at nucleotide position 55. The leucine at codon 19 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
