NM_002769.5(PRSS1):c.559G>A (p.Gly187Ser) was classified as Uncertain significance for Hereditary pancreatitis by Ambry Genetics, citing Ambry Variant Classification Scheme 2023. This variant lies in the PRSS1 gene (transcript NM_002769.5) at coding-DNA position 559, where G is replaced by A; at the protein level this means replaces glycine at residue 187 with serine — a missense variant. Submitter rationale: The p.G187S variant (also known as c.559G>A), located in coding exon 4 of the PRSS1 gene, results from a G to A substitution at nucleotide position 559. The glycine at codon 187 is replaced by serine, an amino acid with some similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6,503 samples (13,006 alleles) with coverage at this position. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this variant remains unclear.
