Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001369.3(DNAH5):c.5578A>G (p.Asn1860Asp), citing Ambry Variant Classification Scheme 2023: The p.N1860D variant (also known as c.5578A>G), located in coding exon 34 of the DNAH5 gene, results from an A to G substitution at nucleotide position 5578. The asparagine at codon 1860 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
