Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_001999.4(FBN2):c.5552T>C (p.Ile1851Thr), citing Ambry Variant Classification Scheme 2023: The p.I1851T variant (also known as c.5552T>C), located in coding exon 44 of the FBN2 gene, results from a T to C substitution at nucleotide position 5552. The isoleucine at codon 1851 is replaced by threonine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
