Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.5495T>C (p.Leu1832Pro), citing Ambry Variant Classification Scheme 2023: The p.L1832P variant (also known as c.5495T>C), located in coding exon 40 of the POLE gene, results from a T to C substitution at nucleotide position 5495. The leucine at codon 1832 is replaced by proline, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
