Uncertain significance — the classification assigned by Ambry Genetics to NM_052947.4(ALPK2):c.5467A>G (p.Lys1823Glu), citing Ambry Variant Classification Scheme 2023: The p.K1823E variant (also known as c.5467A>G), located in coding exon 5 of the ALPK2 gene, results from an A to G substitution at nucleotide position 5467. The lysine at codon 1823 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
