NM_006231.4(POLE):c.541G>T (p.Asp181Tyr) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D181Y variant (also known as c.541G>T), located in coding exon 6 of the POLE gene, results from a G to T substitution at nucleotide position 541. The aspartic acid at codon 181 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
