Uncertain significance — the classification assigned by Ambry Genetics to NM_003628.6(PKP4):c.53G>T (p.Arg18Leu), citing Ambry Variant Classification Scheme 2023: The p.R18L variant (also known as c.53G>T), located in coding exon 1 of the PKP4 gene, results from a G to T substitution at nucleotide position 53. The arginine at codon 18 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
