Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_017617.5(NOTCH1):c.5394G>T (p.Lys1798Asn), citing Ambry Variant Classification Scheme 2023: The p.K1798N variant (also known as c.5394G>T), located in coding exon 29 of the NOTCH1 gene, results from a G to T substitution at nucleotide position 5394. The lysine at codon 1798 is replaced by asparagine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
