NM_013372.7(GREM1):c.536T>C (p.Ile179Thr) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.I179T variant (also known as c.536T>C), located in coding exon 1 of the GREM1 gene, results from a T to C substitution at nucleotide position 536. The isoleucine at codon 179 is replaced by threonine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
