Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_002691.4(POLD1):c.535G>C (p.Gly179Arg), citing Ambry Variant Classification Scheme 2023: The p.G179R variant (also known as c.535G>C), located in coding exon 4 of the POLD1 gene, results from a G to C substitution at nucleotide position 535. The glycine at codon 179 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
