NM_001105206.3(LAMA4):c.5375G>A (p.Cys1792Tyr) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.C1785Y variant (also known as c.5354G>A), located in coding exon 38 of the LAMA4 gene, results from a G to A substitution at nucleotide position 5354. The cysteine at codon 1785 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
