Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000363.5(TNNI3):c.534G>T (p.Lys178Asn), citing Ambry Variant Classification Scheme 2023: The p.K178N variant (also known as c.534G>T), located in coding exon 7 of the TNNI3 gene, results from a G to T substitution at nucleotide position 534. The lysine at codon 178 is replaced by asparagine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
