Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.5309T>A (p.Ile1770Asn), citing Ambry Variant Classification Scheme 2023: The p.I1770N variant (also known as c.5309T>A), located in coding exon 39 of the POLE gene, results from a T to A substitution at nucleotide position 5309. The isoleucine at codon 1770 is replaced by asparagine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
