Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001277115.2(DNAH11):c.5230A>C (p.Thr1744Pro), citing Ambry Variant Classification Scheme 2023: The p.T1744P variant (also known as c.5230A>C), located in coding exon 30 of the DNAH11 gene, results from an A to C substitution at nucleotide position 5230. The threonine at codon 1744 is replaced by proline, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
