Uncertain significance — the classification assigned by Ambry Genetics to NM_001386125.1(OBSCN):c.11458G>A (p.Val3820Met), citing Ambry Variant Classification Scheme 2023: The p.V3391M variant (also known as c.10171G>A), located in coding exon 38 of the OBSCN gene, results from a G to A substitution at nucleotide position 10171. The valine at codon 3391 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
