This variant lies in the NF1 gene (transcript NM_001042492.3) at the canonical splice donor site of the intron immediately after coding-DNA position 5268, where T is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: The c.5205+2T>A intronic variant results from a T to A substitution two nucleotides after coding exon 36 in the NF1 gene. This nucleotide position is well conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice donor site; however, direct evidence is insufficient at this time (Ambry internal data).
