NM_006231.4(POLE):c.5188G>C (p.Asp1730His) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D1730H variant (also known as c.5188G>C), located in coding exon 39 of the POLE gene, results from a G to C substitution at nucleotide position 5188. The aspartic acid at codon 1730 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
