NM_000719.7(CACNA1C):c.5174A>T (p.Gln1725Leu) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q1725L variant (also known as c.5174A>T), located in coding exon 42 of the CACNA1C gene, results from an A to T substitution at nucleotide position 5174. The glutamine at codon 1725 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
