NM_001365951.3(KIF1B):c.5311T>G (p.Cys1771Gly) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.C1725G variant (also known as c.5173T>G), located in coding exon 45 of the KIF1B gene, results from a T to G substitution at nucleotide position 5173. The cysteine at codon 1725 is replaced by glycine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
