Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001277115.2(DNAH11):c.11956G>A (p.Val3986Ile), citing Ambry Variant Classification Scheme 2023: The p.V3986I variant (also known as c.11956G>A), located in coding exon 73 of the DNAH11 gene, results from a G to A substitution at nucleotide position 11956. The valine at codon 3986 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
