Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_006514.4(SCN10A):c.5149T>C (p.Tyr1717His), citing Ambry Variant Classification Scheme 2023: The p.Y1717H variant (also known as c.5149T>C), located in coding exon 27 of the SCN10A gene, results from a T to C substitution at nucleotide position 5149. The tyrosine at codon 1717 is replaced by histidine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
