NM_022051.3(EGLN1):c.511G>C (p.Asp171His) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D171H variant (also known as c.511G>C), located in coding exon 1 of the EGLN1 gene, results from a G to C substitution at nucleotide position 511. The aspartic acid at codon 171 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
