NM_177438.3(DICER1):c.5075A>T (p.Tyr1692Phe) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Y1692F variant (also known as c.5075A>T), located in coding exon 22 of the DICER1 gene, results from an A to T substitution at nucleotide position 5075. The tyrosine at codon 1692 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
