NM_001105206.3(LAMA4):c.5080G>A (p.Glu1694Lys) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E1687K variant (also known as c.5059G>A), located in coding exon 35 of the LAMA4 gene, results from a G to A substitution at nucleotide position 5059. The glutamic acid at codon 1687 is replaced by lysine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
