NM_000093.5(COL5A1):c.5053AAG[1] (p.Lys1686del) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.5056_5058delAAG variant (also known as p.K1686del) is located in coding exon 63 of the COL5A1 gene. This variant results from an in-frame AAG deletion at nucleotide positions 5056 to 5058. This results in the in-frame deletion of a lysine at codon 1686. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
