NM_001042492.3(NF1):c.5110A>G (p.Lys1704Glu) was classified as Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K1683E variant (also known as c.5047A>G), located in coding exon 36 of the NF1 gene, results from an A to G substitution at nucleotide position 5047. The lysine at codon 1683 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
