Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_053025.4(MYLK):c.5041G>A (p.Asp1681Asn), citing Ambry Variant Classification Scheme 2023: The p.D1681N variant (also known as c.5041G>A), located in coding exon 27 of the MYLK gene, results from a G to A substitution at nucleotide position 5041. The aspartic acid at codon 1681 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
