NM_001378454.1(ALMS1):c.5024A>G (p.Tyr1675Cys) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Y1676C variant (also known as c.5027A>G), located in coding exon 8 of the ALMS1 gene, results from an A to G substitution at nucleotide position 5027. The tyrosine at codon 1676 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
