Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001184.4(ATR):c.5007T>G (p.Ile1669Met), citing Ambry Variant Classification Scheme 2023: The p.I1669M variant (also known as c.5007T>G), located in coding exon 28 of the ATR gene, results from a T to G substitution at nucleotide position 5007. The isoleucine at codon 1669 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
