NM_144573.4(NEXN):c.1189C>T (p.Arg397Trp) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R397W variant (also known as c.1189C>T), located in coding exon 9 of the NEXN gene, results from a C to T substitution at nucleotide position 1189. The arginine at codon 397 is replaced by tryptophan, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
