Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001267550.2(TTN):c.76339G>C (p.Val25447Leu), citing Ambry Variant Classification Scheme 2023: The p.V16382L variant (also known as c.49144G>C), located in coding exon 153 of the TTN gene, results from a G to C substitution at nucleotide position 49144. The valine at codon 16382 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be benign and unknown by PolyPhen and SIFT in silico analyses, respectively. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
