This variant lies in the BRCA2 gene (transcript NM_000059.4) at coding-DNA position 4895, where G is replaced by A; at the protein level this means replaces serine at residue 1632 with asparagine — a missense variant. Submitter rationale: The p.S1632N variant (also known as c.4895G>A), located in coding exon 10 of the BRCA2 gene, results from a G to A substitution at nucleotide position 4895. The serine at codon 1632 is replaced by asparagine, an amino acid with highly similar properties. This alteration has been reported in a cohort of Korean breast cancer patients (Kim HN et al. Chonnam Med J, 2019 May;55:99-103). This amino acid position is not well conserved in available vertebrate species, and asparagine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
