NM_001277115.2(DNAH11):c.10166-1G>C was classified as Likely pathogenic for Primary ciliary dyskinesia by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.10166-1G>C intronic variant results from a G to C substitution one nucleotide upstream from coding exon 63 of the DNAH11 gene. This nucleotide position is highly conserved in available vertebrate species. Based on the ESEfinder in silico model, this alteration is predicted to abolish the native splice acceptor site; however experimental evidence is not currently available. Alterations that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay. As such, this alteration is classified as likely pathogenic.
