Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.4874A>G (p.Gln1625Arg), citing Ambry Variant Classification Scheme 2023: The p.Q1625R variant (also known as c.4874A>G), located in coding exon 37 of the POLE gene, results from an A to G substitution at nucleotide position 4874. The glutamine at codon 1625 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
