Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001003800.2(BICD2):c.479G>T (p.Arg160Leu), citing Ambry Variant Classification Scheme 2023: The p.R160L variant (also known as c.479G>T), located in coding exon 3 of the BICD2 gene, results from a G to T substitution at nucleotide position 479. The arginine at codon 160 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
