NM_001365951.3(KIF1B):c.4934C>G (p.Ser1645Cys) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S1599C variant (also known as c.4796C>G), located in coding exon 42 of the KIF1B gene, results from a C to G substitution at nucleotide position 4796. The serine at codon 1599 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
