NM_005359.6(SMAD4):c.478G>C (p.Asp160His) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome; Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D160H variant (also known as c.478G>C), located in coding exon 4 of the SMAD4 gene, results from a G to C substitution at nucleotide position 478. The aspartic acid at codon 160 is replaced by histidine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
