NM_058216.3(RAD51C):c.476A>T (p.Asp159Val) was classified as Likely pathogenic for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D159V variant (also known as c.476A>T), located in coding exon 3 of the RAD51C gene, results from an A to T substitution at nucleotide position 476. The aspartic acid at codon 159 is replaced by valine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In a homology-directed DNA repair (HDR) assay, this alteration showed a functionally abnormal read-out (Olvera-Le&oacute;n R et al Cell 2024 Oct;187(20):5719-5734.e19). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the majority of available evidence to date, this variant is likely to be pathogenic.
