Uncertain significance for Hereditary pancreatitis — the classification assigned by Ambry Genetics to NM_007272.3(CTRC):c.475G>A (p.Gly159Ser), citing Ambry Variant Classification Scheme 2023: The p.G159S variant (also known as c.475G>A), located in coding exon 5 of the CTRC gene, results from a G to A substitution at nucleotide position 475. The glycine at codon 159 is replaced by serine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
