Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001378454.1(ALMS1):c.4742A>G (p.Tyr1581Cys), citing Ambry Variant Classification Scheme 2023: The p.Y1582C variant (also known as c.4745A>G), located in coding exon 8 of the ALMS1 gene, results from an A to G substitution at nucleotide position 4745. The tyrosine at codon 1582 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
