Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001018005.2(TPM1):c.470A>C (p.Asp157Ala), citing Ambry Variant Classification Scheme 2023: The p.D157A variant (also known as c.470A>C), located in coding exon 4 of the TPM1 gene, results from an A to C substitution at nucleotide position 470. The aspartic acid at codon 157 is replaced by alanine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
